1、T淋巴瘤侵袭转移诱导因子1与胃癌细胞侵袭移行能力的关系作者:朱金明余佩武李翠芳吴淼【摘要】目的检测胃癌细胞中T淋巴瘤侵袭转移诱导因子1(Tlymphomainvasionandmetastasisinducingfactor1,Tiam1)的表达及其与肿瘤侵袭、移行能力的关系,同时观察胃癌细胞在形态学方面的变化。方法采用层黏连蛋白黏附法,由胃癌MKN45细胞株(M0)中筛选获得高(MH)、低(ML)黏附亚株。应用流式细胞仪检测Tiam1在胃癌细胞中的表达,以Boyden小室法测定M0、ML、MH细胞的体外侵袭、移行能力,并分析其与Tiam1表达间的关系,同时采用苏木精伊红
2、及细胞骨架蛋白染色、扫描电镜技术观察胃癌细胞形态。结果MH细胞的体外侵袭、移行能力及Tiam1表达均强于M0、ML细胞(P<0.05),且Tiam1表达与胃癌细胞体外侵袭、移行能力呈正相关(r=0.997和1.000);同时与M0、ML相比,MH胞体伸展,表面突起增多,片状伪足宽厚,丝状伪足细长而密集,异型性明显,且细胞骨架结构较紊乱,斑点状肌动蛋白小体增多。但M0、ML细胞间差异无统计学意义(P>0.05)。结论Tiam1表达水平升高可能促进胃癌细胞侵袭、移行。这或许是通过调整胃癌细胞骨架结构重组,增强其变形、游走能力而实现的。11【关键词】胃癌;T淋巴瘤侵袭转
