2、COX1andCOX2inhumanastrocytomatissueandnormalbraintissue.METHODS:Reversetranscriptasepolymerasechainreaction(RTPCR)andimmunohistochemistry(SP)wereappliedtodetectthemRNAandproteinexpressionsofCOX1andCOX2inhumanastrocytomatissueandnormalbraintissue.FurthemoretherelationshipbetweenCOXexpressionl
3、evelsandcancerpathologicalgradeswasanalyzed.RESULTS:UsingRTPCR,all20freshspecimensfromhumanastrocytomaswereshowntoexpressCOX2mRNA,whilenopositive12expressionofCOX2mRNAwasfoundin8specimensofnormalbraintissues;therewasasignificantdifferencebetweenthetwo(P<0.0001).ButCOX1mRNAwasexpressedcons
4、titutionallyinall20ofthefreshcancertissuesand8samplesofnormalbraintissues.TheimmunohistochemicalresultshowedthatCOX2proteinwasmainlylocalizedatcytoplasmofmalignantcells(positiveratewas72%),butwasnotexpressedinnormalbraintissues.Incontrast,COX1proteinwasmainlylocalizedatbraincellsinnormalbrai
5、ntissue(positiverate100%),butwasnotexpressedinhumanastrocytomas.In50paraffinembeddedspecimensofhumanastrocytomas,theintensityofCOX2expressionwasassociatedwithtumorgradeandstage.ThelevelofCOX2expressionwashigheringradeⅢ,ⅣtumorsthaningradeⅠ,Ⅱtumors.CONCLUSION:TheexpressionofCOX2mRNAandproteini

7、】目的:通过检测环氧化酶1(COX1)和环氧化酶2(COX2)在人脑星形细胞瘤、正常脑组织中的表达,探讨COX在人脑星形细胞瘤发生发展中的作用.方法:应用逆转录聚合酶链反应(RTPCR)和免疫组化SP法检测人脑星形细胞瘤、正常脑组织中COX1,COX2mRNA和蛋白的表达及其与肿瘤恶性程度、病理级别的关系.结果:RTPCR检测20例人脑星形细胞瘤组织COX2mRNA均阳性表达,正常脑组织中未见表达,两者有显著性差异(P<0.0001);COX1mRNA在20例肿瘤标本及正常脑组织中均有
