1、56一现代中药研究与实践2014年第28卷第1期一ChinMedJResPrac,2014Feb.Vo1.28No.1姜黄素包合物胃内漂浮微球的研制张晓冲,廖启元,翟光喜(1.安徽中医药高等专科学校,安徽芜湖241002;2.山东大学药学院,山东济南250000)摘要:目的制备姜黄素胃内漂浮微球。方法为增加姜黄素的水溶性,先制备了姜黄素的羟丙、基一13一环糊精包合物,通过正交试验确定了最优处方。以乙基纤维素(粘度45)为载体材料,采用液中干燥法制备胃内漂浮微球,在单因素考察的基础上,采用Plackett-Burman筛选显著因素,结合响应面法优化微球制备工艺。对
2、微球的外观形态、流动性、堆密度、粒径分布、漂浮率、包封率、载药量、体外累积释药百分率等进行考察。结果最佳工艺为:EC/HP—B—cD—cuR(质量比)为7.6:1,温度44-3℃,转速801r/rain,油相和药物所在有机相体积比2:l,乳化剂用量为O.030g/mL,EC的浓度为O.020g/mL。结论姜黄素微球形态圆整,大小均匀,无粘连;平均粒径为680.3m,平均包封率与载药量分别为64.6%和4.65%,在人工胃液中有良好的漂浮性;漂浮微球体外释药符合一级动力学方程,呈现明显的缓释性。关键词:姜黄素;漂浮微球;响应面;包合物中图分类号:R944.16文献
