4、sreviewoutlinesoursearchforthemechanismcausingtheearlylossofisletsympatheticnervesinautoimmunediabetes.Sinceourpreviousworkhasdocumentedtheimportanceofautonomicstimulationofglucagonsecretionduringhypoglycaemia,thelossofthesenervesmaycontributetotheknownimpai
5、rmentofthisspecificglucagonresponseearlyinhumantype1diabetes.Wethereforebrieflyreviewthecontributionthatautonomicactivation,andsympatheticneuralactivationinparticular,makestothesubsequentglucagonresponsetohypoglycaemia.Wealsodetailevidencethatanimalmodelsofa
6、utoimmunediabetesmimicboththeearlylossofisletsympatheticnervesandtheimpairedglucagonresponseseeninhumantype1diabetes.Usingdatafromtheseanimalmodels,weexaminemechanismsbywhichthislossofisletnervescouldoccur.Weprovideevidencethatitisnotduetodiabetichyperglycae
7、mia,butitisrelatedtothelymphocyticinfiltrationoftheislet.Ablatingthep75neurotrophinreceptor,whichispresentonsympatheticaxons,preventseSIN,but,interestingly,notdiabetes.Thus,weappeartohaveseparatedtheimmune-relatedlossofisletsympatheticnervesfromtheimmune-med
