1、www.zgys.org研究论文不同纯化工艺对知母皂苷水提液的影响1211221梁艳妮范瑞韬王征刘世军葛秋萍桑盈盈唐志书(1.陕西中医药大学/陕西省中药资源产业化协同创新中心陕西省中药基础与新药研究重点实验室陕西咸阳712083;2.咸阳市实验中学)摘要目的:考察膜分离与大孔树脂吸附技术对知母皂苷水提取液的纯化作用。方法:采取煎煮2次,10倍量水,每次120min的水煎煮工艺提取知母皂苷母液,分别采用不同型号的大孔树脂(HP20、HPD600、D101、AB8)吸附及膜分离(陶瓷微滤膜0.8,0.05μm和中空纤维超滤膜50,10,6kDa)技术纯化知
2、母水提液,测定其电导率、黏度、浊度等物理化学参数及蛋白质、多糖、总皂苷等物质含量。结果:知母皂苷水提取液经不同膜过滤后黏度和浊度减小,pH增大,电导率无规律性改变,微滤膜比超滤膜对药液的澄清效果好;孔径越小,截留相对分子质量越小,蛋白质截留率越高,皂苷透过率也越高,多糖含量差异不大,与理化参数的改变一致。微滤膜澄清药液效果最佳,截留相对分子质量6kDa的超滤膜更适合皂苷成分的富集。最适合知母皂苷水提液纯化的大孔树脂为HPD600。结论:膜组件的选择对于知母皂苷的分离纯化显得尤为重要。理化参数的改变与大分子物质的含量减小有着明显的对应关系,超滤膜比微滤膜更适
