BPHMiceandRatsModelsZuoTing(Pharmacology)DirectedbyMiaoMingsanAbstractObjective:ToobservetheeffectofWujiBaifengPellettobenignprostatichyperplasia(BPH)inmiceandratsmodels.Throughmeasuretheprostateweight,prostateexponent,determinethele
2、veloftestosterone(T)、estradiol(E2)andepidermalgrowthfactor(EGF)contentintheanimalmodels,andobservethemorphologicalchangeofthemiceandrats’prostate,thymus,renalandspleen.ToevaluatecurativeeffectandinitiallyrevealclarifytherapeuticactionofWujiBaifengPelletrestrainingBPH.ofmiceandrat
3、smodels.Methods:Bysubcutaneousinjectingtestosteronepropionate5mg/kgincastratedmiceeverydaytocopytheBPHmicemodel.Threedayslaterfrommicecastrated,respectivelydrenchthesuspl.oflongbishucaps(0.45g/kg,fiftytimesofclinicdosage),thesuspl.ofwujibaifengpellet(9g/kg、4.5g/kg、2.25g/kg,2ml/10
4、0g)andthesamedosagesalineintheblankgroupandmodelgroup,onceadayforthreeweeks.Twohourafterlastadministration,takebloodfromeyesockets,putthemicetodeath,observethechangesofprostateweight,prostateexponent,measuretheT、E2contentinbloodserum,andobservethemorphologicalchangeofthemiceandra
5、ts’prostate,thymus,renalandspleenineachgroup.Bysubcutaneousinjectingtestosteronepropionate4mg/kg/dincastratedmiceeverydaytocopytheBPHmicemodel.Sevendayslaterfrommicecastrated,respectivelydrenchthesuspl.oflongbishucaps(0.3g/kg,tentimesofclinicdosage),thesuspl.ofwujibaifengpellet(6
6、g/kg、3g/kg、1.5g/kg,2ml/100g)andthesamedosagesalineintheblankgroupandmodelgroup,onceadayfor30days.Twohourafterlastadministration,theratswereputtodeath,observedthechangesofprostateweight,prostateexponent,histologyunderlightmicroscopeofeachgroup,measuredtheT、E2、EGFcontentofprostatec
