3、cedbyRNAinterference(RNAi)inwhichmaletickswereinjectedwithvarisindsRNAandthenallowedtofeedandacquireA.marginaleinfectiononanexperimentally-infectedcalf.Silencingexpressionofvarisininhemocytes,midgutsandsalivaryglandswasconfirmedbyrealtimeRT-PCR.Weexpe
4、ctedthatsilencingofvarisinwouldincreaseA.marginaleinfectionsinticks,buttheresultsdemonstratedthatbacterialnumbers,asdeterminedbyanA.marginalemsp4quantitativePCR,weresignificantlyreducedinthevarisin-silencedticks.Furthermore,coloniesofA.marginaleintick
5、susedforRNAiweremorphologicallyabnormalfromthoseseeninelutionbufferinjectedcontrolticks.ThecolonyshapewasirregularandinsomecasestheA.marginaleappearedtobefreeinthecytoplasmofmidgutcells.Sometickswerefoundtobesystemicallyinfectedwithamicrobethatmayhave
