1、面北雇盐亏搪2011,20(5):111—116ActaAgriculturaeBorealioccidentalisSinica水稻土氨氧化细菌多样性的RFLP分析李惠民。,程林,王保莉,曲东。(1.西北农林科技大学生命科学学院,陕西杨凌712100;2.商洛职业技术学院,陕西商洛7260003.西北农林科技大学资源环境学院,陕西杨凌712100)摘要:提取苗期水稻根际土和非根际土土样微生物总DNA,采用氨氧化细菌特异性引物(Eub338,Nso1225)扩增16SrDNA基因片段,分别建立水稻根际土(G)和非根际土(F)氨氧化细菌克隆文库。用限制性内切酶HhaI/RsaI进行PC
2、R—RFLP分型,分别得到110和105个酶切类型。多样性指数和优势细菌聚类比对结果显示,土壤氨氧化细菌群落结构指数H、D和-,为非根际土稍大于根际土,表明非根际土中氨氧化细菌种群比根际土略多。指数H⋯和d为根际土大于非根际土,表明根际土氨氧化细菌数量相对多于非根际土。根际土有比非根际土更明显的优势种群出现。序列分析表明,水稻根际土氨氧化细菌的优势种群主要属于亚硝化螺菌属,亚硝化单胞菌属,p一变形菌亚门,碱菌属。供试水稻根际土和非根际土氨氧化细菌多样性和群落结构发生了改变。关键词:水稻土;氨氧化细菌;多样性;RFIP中图分类号:S511文献标志码:A文章编号:1004—1389(20
