2、生,为刺槐生产及育种提供参考依据。【方法】以刺槐花粉为试材,使用培养基萌发试验法测定其生活力,并研究不同浓度ca”、不同贮藏条件及时间对刺槐花粉萌发的影响。【结果】刺槐花粉在琼脂8.0g/L
3、+蔗糖20.0mg/L+硼酸2.0mg/L的培养基中萌发率高达30.25%;适当浓度的ca2_能促进花粉萌发和花粉管伸长,当caz榷度为1.6x10。mg/L时,花粉萌发率高达40.25%,花粉管伸长最快;适宜刺槐花粉贮藏的条件依次为名o℃、4℃、一23℃和常温;花粉在名o℃贮藏32d后,萌发率仍达15.30%。【结论】适合刺槐花粉萌发的培养基为:琼脂8.0g/L+蔗糖20.0mg/L+硼酸2.0mg/L;培养基中添加低浓度外源钙能促进刺槐花粉萌发和花粉管生长;于超低温真空冷冻条件下贮藏的刺槐花粉保持活力时间更长。关键词:刺槐;花粉活力;贮藏条件;培养基萌发法
