3、imentaltechniquerequiresallenzyme-catalyzedreactionstobeinstanta-neouslystoppedintheintacttissuesothatthemetabolicintermediatesdonotundergochange.Thisobjectiveisaccomplishedbyrapidlycompressingthetissuebetweenlargealuminumplatescooledwithliquidnitrogen(190C),aprocesscalled
4、freeze-clamping.Afterfreezing,whichstopsenzymeactioninstantly,thetissueispowderedandtheenzymesareinactivatedbyprecipitationwithperchlo-ricacid.Theprecipitateisremovedbycentrifugation,andtheclearsupernatantextractisanalyzedformetabolites.Tocalculateintracellularconcentrations
