1、重庆大学硕士学位论文英文摘要AbstractThecellelasticpropertyisoneimportantparameteronthecellbiomechanicsresearch,thestructureandphysiologicalfunctionsofcellsarecloselyrelatedwiththegrowthenvironmentofcell,andthephysicalchangingofmicroenvironmentofcellcannotonlycausethecellelasticitychangedbutalsot
2、heexpressionofintegrinandoutsideofthecellorientation.Theycanaffecttheinteractionsbetweenintegrinandtheirligands.Thechitosan/polyacrylamidehydrogelisavariablesubstratematerial,andHepG2cellsareplantedondifferentrigidityofchitosan/polyacrylamidehydrogensubstrates.After48hours,morpholo
3、gicalcharacteristicsofHepG2cell(suchasspreadarea,long/shortaxisratio)aremeasured,andthecytoskeletonisobservedbyFITC-phalloidin,thentheexpressionofintegrinα6β1isdetectedusingFCM;thesurfacetopography;cellelasticityofHepG2indifferentrigiditysubstrateandtheinteractionsbetweenintegrinα6
4、β1andLNunderphysiologicalconditionsaredetectedbyAFM.Themainresultsobtainedinthisstudyasfollows:(1)Therigidityofchitosan/polyacrylamidehydrogelisadjustable,andcanbeusedasbasematerialforcellmechanicsresearch.(2)Thegreaterbasalrigidity,themorespreadareaandlong/shortaxisratioofHepG2cel
5、l.(3)ThesurfacetopographyandcellelasticityofHepG2cellarediscrepantindifferentrigidity.Thelesspseudopodiaextendingfromcelledgeinthehardnessofsubstrateof1000Pa,themorepseudopodiainthecontrolgroup.TheYoung’smodulusofHepG2cellsinthecontrolgroup,3500Paand1000Parigiditysubstrateare2036+1
6、045Pa,714+311Paand803+445Pa.(4)Theadhesionproportionofcellisdifferentineachrigidity,theinteractionforcebetweenintegrinα6β1andligand(LN)are19±7pN、37.5±15.8pNand38.85±19.7pN.(5)ThecytoskeletoncolorationsdetectedthatthecytoskeletonofHepG2isclearandthroughoutthecellincontrolgroup,while
