2、(5’.CGCAAGCTTa盯AATTACACCAClAGGCA.3’);以pT哂以C为模板进行PCR扩增,扩增产物克隆至pMDl8.T中,构建重组质粒pMDl8.T哂谢C;双酶切后将s以C基因的全编码序列克隆至pMG36e的劢口I和砌dIII之间的多克隆位点上,构建表达重组子pMG36e哂DdC;然后通过优化的电转化条件(电压2.5kV,电阻100Q,电容2和F),将pMG36e心DdC转化至干酪乳杆菌L反c口sd,并对重组菌株进行质粒抽提、PCR鉴定、酶切鉴定。结果表明成功地构建了重组干酪乳杆
