1、上海交通大学硕士学位论文蓝百合快速繁殖技术的研究姓名:范现丽申请学位级别:硕士专业:园林植物与观赏园艺指导教师:申晓辉20090101胚后期可产生次生胚,最后都能形成正常植株。诱导胚状体的最佳培养基为MS+6-BA0.1mg/L+蔗糖45g/L,每g细胞鲜重可产生722个胚状体,比Sakaeetal.以叶片为外植体,在MS+1mg/LPIC诱导产生的胚状体数量(476个)提高了一半多(51.68%)。7.蓝百合最佳生根培养基为1/2MS+IBA0.l5mg/L,根系数量多,长势良好,健康而且粗壮。当组培苗和体胚苗大约高2-3c
2、m时,选择健壮的苗,即可移栽。移栽后两周内保持适宜的光照和湿度,成活率可达98.5%。8.通过蓝百合由增殖到成熟胚诱导阶段的细胞显微结构观测,对比禾本科植物发生方式研究可知,本项研究首次证明了蓝百合在同一种培养物中存在两种体细胞胚胎发生方式,即胚胎发生方式和器官发生方式。9.采用蓝百合根茎顶端为外植体进行愈伤组织的诱导,其诱导率比花梗诱导率提高40%,并获得了成活率高达98.5%的体细胞再生植株,为我国获得具有自主知识产权的蓝百合快速繁殖产业化生产技术奠定了理论和实践基础。关键词:‘蓝色大花’蓝百合组织培养体细胞胚胎发生组织学

5、cation.Theresultswereasthefollowed:1.Seedgerminationrateofpretreatmentwithremovingseedwingandaddingdetergentwas42.2%andhigherthannon-pretreatment.2.TheoptimizingmediumforinducingcallusfrompedicelwasMS+6-BA1.5mg/L+Citricacid0.1mg/Landtheinductionratewas13.33%.However
6、,theinductionratewas56.7%fromadventitiousbudsontheoptimizingmediumcontainingMS+6-BA1.5mg/L+IBA0.2mg/L.Pedicelasexplantsislimitedseriouslybysamplingtime,soitcouldnotachievetherequirementsofindustrialization.3.Caudexes,leavesandpedicelswereusedasexplantsinsomaticembry
