1、万方数据2.暗纹东方纯卵子磷酸酶等生化指标分析与卵质评估用生化分析方法对暗纹东方纯未受精卵子基本成分、磷酸酶活力、苹果酸脱氢酶活力、唾液酸、氨基酸含量进行分析,同时用GC—MS方法对脂肪酸组成及相对含量进行检测,并对各物质变化规律与其孵化率的相关性进行分析。结果显示,酸性磷酸酶活力、唾液酸含量与孵化率显著相关(P<0.05,R12=0.501,R22=0.406),碱性磷酸酶与孵化率显著负相关(P2、R22=0.710),并呈现先递增后递减的变化趋势;n.3脂肪酸、DHA与EPA之比均与孵化率存在显著相关(尸<0.05,R12=0.509,R22=0。722,R32=0.550)。结果表明,磷酸酶、唾液酸、脂肪酸等物质变化规律与卵子孵化率存在相关性,且ACP活力值在[68.045,141.038]范围时,AKP活力值在【2.298,7.24I]范围时,SA含量在[9.08l,19.973]范围时,C14:0含量在【0.485,0.6581范围时,ARA含量在[1.371,2.498]范围时,DHA含量在[13.838,18.469]范围时,均可作为卵子孵化率不低于503、%的参考指标,据此可在一定程度上反应出暗纹东方纯苗种繁育生产中所用卵子的可孵化价值,亦可作为参考指标对其卵子质量进行初步、客观评价。3.红鳍东方纯卵子生理生化分析与质量评估人为对性成熟亲鱼各推迟一周、两周挤卵,并与正常组作比较,研究其推迟产卵后卵子内磷酸酶、苹果酸脱氢酶、脂肪酸等生化参数变化规律及其与受精率的相关性。结果显示,推迟一周产卵后,B卵子组受精率显著下降(P4、、C两组(P<0.05),总氨基酸含量亦存在相似变化趋势,A、B组显著高于C组(,I<0.05):此外C14:0、C15:0、C16:0、C16:In.7、C18:3n一3、C20:5n.3在总脂中比例均在正常组中最高,并与受精率存在显著正相关(p<0.05);C22:5n.3、DHA:EPA均与受精率呈显著负相关(pcO.05)。结果表明,酸性磷酸酶、苹果酸脱氢酶、总氨基酸及多种脂肪酸可在一定程度反应红鳍东方纯卵子过熟与否,亦可作为参考指标对其卵子质量进行初步评价。关键词:暗纹东方纯,家系构建,卵子,生化指标,质量评估万方数据Large—scalefamilyconst5、ructionofTakifuguobscurusandeggqualityassessmentOiltwokindsofTakiuguAbstractTakifuguobscLI嬲andTakifugurubripeSbelongtoTakifugu,bothofthemareimportanteconimicfishesinChina.Butthemethodoflarge-scalefamilyconstructionofTakififguobscurushash’testablishedyet,alsoasanimportantfactorinfluencingt6、heearlystageoffamilyconstruction,thequalityofTalafuguobscurusandTakifugurubnpeseggsstillhavn’thaveevaluationcriteria.Toestablishthemethodoflarge-scalefamilyconstruction,toinvestigatetherelationshipbetweenthedevelopmentalcapacityandbiochemicalparametersofegginTakifuguobscurus,alsotoinvesti7、gatethevariationofphysiologicalandbiochemicalparametersintheover-ripeningeggsofTakifugurubripesandtheeffectoneggquality,Threepartsofexperimentwerestudiedinthispaper,thedetailsareasfollows:1.FamilyconstructionandearlystandardizedcultivationofTakifuguobscurtisWitllthe
