1、ABSTRACTRecently,therehasbeenarapidgrowthinthedevelopmentofthehighlyselectiveandsensitiveopticalnanobiosensorsbasedongoldnanoparticlesmodifiedfilmsfordiseasediagnosis,drugdiscovery,proteomicsandmolecularbiology.Takingpolyvinylpyrrolidone(PVP)asprotectiveagentandsodiumcitrateas

2、reducingagent,homogeneousgoldsolswerepreparedbychemicalreductioninthispaper.DL-1,4-dithiothreitol(DTT)wasfirstself-assembledonaflatgoldplateelectrodewithasurfaceareaof5mmx5mm,3-mercaptopropionicacidwaschosentobindwithgoldparticstoformmolecularrecognitionbiosensorsincomparisonw

3、iththatofL—cysteine.ThepreparedgoldnanoparticleswerecharacteredwithUV-VisandTEM.Andtheirassemblingprocessesweretestedbycylicvoltammetry.BrokenthroughthetraditionalcatalysisofEDC—NHS,theEDC-DMAPasanicecatalystofinteractionbetweenaminoandcarbonoxylgroupshasbeenexploited.Thenanob

4、iosensor,basedonthegoldnanoparticlesmodifiedwiththeorganicfunctionalgroupswithactivityincreased,exhibiteddistinctopticalpropertiesoflocalizedsurfaceplasmonresonance(LSPR)invisibleabsorptionregion.Themaximumadsorptionamountofc-mycmonoclonalantibodymoleculeonthemodifiedLSPRnanob

5、iosensorwiththenovelorganicfunctionalnanofilmcouldreach2.4pg/mL.TheLSPRbiosensorimmobilizedwithc-mycmonoclonalantibodycouldbeusedforrecognitionofoncogenebiomoleculessuchasMYCrecombinantprotein(P01)withalowlinearresponserangeof6.5x10一一1.3I.tg/mLandadetectionlimitof6.5x10一I-tg/m
