1、苦参碱对溃疡性结肠炎大鼠肠粘膜细胞因子和自由基的影响熊永爱1赵强强1王淼1黄勤挽1谢兴亮1杨明*1(1成都中医药大学药学院中药材标准化教育部重点实验室四川成都611137)摘要目的探究苦参碱对溃疡性结肠炎大鼠肠粘膜细胞因子和自由基的影响机制。方法利用三硝基苯磺酸(TNBS)复制大鼠溃疡性结肠炎模型,灌胃给予苦参碱溶液进行治疗。治疗结束后,剖取结肠,检测溃疡性结肠粘膜细胞中IL-1β、TNF-α、IL-4、IL-10等一系列细胞因子的水平;检测结肠粘膜细胞中SOD、MDA水平。另取部分结肠做组织显微镜检查,并对组织损伤评分进行数理统计。结果1)与模型组相比,苦参碱和柳氮磺胺吡啶均
2、能极显著降低IL-1β水平(p<0.01);显著降低TNF-α水平(p<0.01);极显著降低IL-4水平(p<0.01);极显著降低IL-10水平(p<0.01);2)与模型组相比,苦参碱和柳氮磺胺吡啶均能极显著升高结肠粘膜细胞SOD水平(P<0.01);极显著降低MDA水平(P<0.01);3)苦参碱和柳氮磺胺吡啶均能促进溃疡面愈合,减少病灶部位炎细胞浸润,水肿及纤维化。结论苦参碱能明显抵抗溃疡性结肠炎炎性反应,增强机体免疫,通过调节肠粘膜细胞因子失衡和抑制粘膜细胞氧自由基的产生和抗氧化功能干预了UC发病过程。关键词苦参碱溃疡性结肠炎肠粘膜细胞因子自由基机制Abstract
