1、基于免疫层析技术的时间分辨荧光免疫分析仪研究第30卷第6期2009年6月仪器仪表ChineseJournalofScientificInstrumentVo1.30No.6Jun.2009基于免疫层析技术的时间分辨荧光免疫分析仪研究任冰强.-.黄立华r,黄惠杰(1中国科学院上海光学精密机械研究所上海201800;2中国科学院研究生院北京100039)摘要:基于时间分辨荧光免疫分析和免疫层析技术的原理,研制了一种新型的时间分辨荧光免疫分析仪.与现有的时间分辨荧光分析仪不同,它所检测的是以Eu"离子及其螯合物为标记物的免疫层析试纸条.通
2、过测量免疫反应后试纸条检测带上Eu"螯合物的含量,参照标准浓度曲线就可以定量得到待测样品溶液的浓度.实验表明,仪器的最低检测浓度可达到ng/mL量级,测量的相对标准偏差小于3%,在5~10ng/mL浓度范围内具有良好的线性响应特性,相关系数R=0.9997.关键词:传感器技术;标记免疫学;时间分辨荧光免疫分析仪;免疫层析技术中图分类号:TP212.3文献标识码:A国家标准学科分类代码:460.4035Time-resolvedfluorometerbasedonimmunochromatographyRenBingqiang一,Hu

4、theoth—ertime—resolvedfluorometers,itteststheimmunochromatographicstripthatislabeledwithlanthanideionsandtheirchelates.Thisinstrumentcanprovidearapid,quantitativemeasurementofanalytespresentedinthesampleswithoutanywashingstepsanditcanbeusedtocarryoutpoint-of—caretest(P
5、OCT).Theinstrumentiscomposedofaspe—cificopticalsensor,ascanningstage,asignalprocessingsystemandacomputercontrolsystem.AUVLEDwithpickwavelengthof345nmisusedastheexcitinglightsourcethatismodulatedat1kHz.ThetightfromtheLEDisfocusedontheteststripbyafocusinglensgroup.IftheE
6、u"chelatesarepresentinthesamples,thefluorescencewithpickwavelengthof613nmwillbeexited.AfteradelayofsomemLcroseconds,thefluorescencewillbecollect—edbytheopticalsensorandconvertedintoelectronicsignalbyaphotomuhipliertube(PMT).Theconcentrationofthesamplecanbecalculatedacc
