2、养法,设置培养基组分、pH和温度等8个影响因子的梯度值进行逐步迭代优化,以缺素培养为对照(CK),探讨其花粉萌发效果。【结果】培养基各组分对宫粉羊蹄甲花粉离体萌发的影响效果迥异,其中H3BO,对花粉萌发起决定性作用,花粉萌发对蔗糖较敏感。宫粉羊蹄甲花粉在添加8.0%蔗糖、10.0%聚乙二醇.6000(PEG.6000)、0.0020%H墨03、0.02%CaCl2·2H20、0.02%MgS04·7H20及0.010%KNO,的液体培养基中,于pH6.8、恒温29℃条件下培养约20min可观察到最佳萌发效果,萌发率均值达91.230%。【结论】宫粉羊蹄甲花粉
