1、温阳活血方上调慢性马兜铃酸肾病大鼠肾组织血管生成素1mRNA表达作者:王红婷,王云满,梁永平,刘育军,彭文【摘要】 目的:观察温阳活血方对慢性马兜铃酸肾病大鼠肾组织血管生成素(angiopoietin,Ang)表达的影响,并探讨其保护作用机制。 方法:将28只雄性SD大鼠随机分为正常对照组、模型组和温阳活血方组。模型组大鼠予关木通水煎液10ml/(kg·d)灌胃,温阳活血方组于模型组基础上,再予温阳活血方30g/(kg·d)灌胃。20周后,采用实时逆转录聚合酶链式反应方法检测3组大鼠肾组织中Angl和Ang2mRNA的表达。 结果:与正常对照组比较,模型组大鼠肾组织Ang1m
2、RNA表达明显减少(P<0.01),Ang2mRNA表达明显升高(P<0.01);与模型组大鼠比较,温阳活血方组大鼠肾组织Ang1mRNA表达升高(P<0.01),Ang2mRNA表达下降,但差异无统计学意义(P>0.05);肾脏病理显示温阳活血方组肾损害较模型组明显减轻。 结论:温阳活血方通过上调Ang1mRNA的表达发挥对慢性马兜铃酸肾病大鼠肾脏保护作用。【关键词】马兜铃酸;温阳活血方;血管生成素类;关木通;大鼠;肾病 Objective:ToinvestigatetheeffectsofWenyangHuoxueRecipe(WRHXR),aco

5、/(kg·d)andCAMdecoction10ml/(kg·d).TheexpressionsofAnglandAng2mRNAsweredetectedbyrealtimereversetranscriptionpolymerasechainreaction(RTPCR)after20weektreatment. Results:Comparedwiththenormalcontrolgroup,theexpressionofAnglmRNAwassignificantlydecreased,andtheexpressionofAng2mRNAwassignifica
6、ntlyincreasedintheCAMgroup(P<0.01).ComparedwiththeCAMgroup,theexpressionofAnglmRNAwasincreasedintheWYHXRtreatedgroup(P<0.01).TheexpressionofAng2mRNAhadnosignificantdifferencebetweentheCAMgroupandtheWYHXRtreatedgroup(P>0.05).RenalpathologyshowedthatrenaldamageinWYHXRtreatedgroupwassi
