1、杨梅素减轻高糖诱导的血管内皮细胞氧化应激损伤梁馨予,张婷,周永,施琳颖,陈军丽,王丽,康超,朱俊东,糜漫天(400038重庆,第三军医大学军事预防医学院营养与食品安全研究中心,重庆市营养与食品安全重点实验室,重庆市医学营养研究中心)[摘要]目的探讨杨梅素对高糖诱导的血管内皮细胞氧化应激损伤的影响。方法体外培养原代人脐静脉内皮细胞(HumanUmbilicalVeinEndothelialCells,HUVECs),分为正常对照组(葡萄糖浓度5.56mmol/L)、甘露醇高渗对照组(5.56mmol/L葡萄糖+27.8mmol/L甘露醇)、高糖处理组(葡萄糖浓
2、度33.36mmol/L)、杨梅素保护组(0.1~10µmo/L杨梅素+33.36mmol/L葡萄糖),处理48h后,CCK-8比色法检测细胞活力,Annexin-FITC/PI双染流式细胞术检测细胞凋亡,同时检测细胞内活性氧(ROS)含量,及培养上清液中丙二醛(MDA)含量、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性。结果高糖处理HUVECs48h后,与正常对照组比较,细胞活力显著降低(P<0.05),细胞凋亡率、ROS及MDA含量均显著增加(P<0.05),而SOD和CAT活性显著下降(P<0.05);杨梅素预处理HUVECs后,可显著抑制高糖引
