1、中国协和医科大学博士学位论文第一节酵母双杂交筛选随机多肽文库研究ErbinPDZ的结合特性摘要:Erbin(ErbB2interactingprotein)可通过其PDZ结构域介导ErbB2定位到上皮细胞基底外侧膜。为了深入研究ErbinPDZ结构域的配体识别特性,本实验以ErbinPDZ为诱饵蛋白,用酵母双杂交系统来筛选新型随机多肽文库。研究发现,除了报道的I类配体-IE/D]一[V/S]一W—V之外,ErbinpDZj2、酸性的E或D也可被识别。实验同时得到了高亲和力的ETwv配体,也发现了低亲和力的ESDV。综合分析所有得到的阳性克降,发现其在c.末端呈现规律。利用规律搜索Swiss.Prot数据库预测ErbinPDZ潜在的天然配体。经过初步验证,得到了4个新的天然配体,为深入研究ErbinPDZ的功能提供了线索。上述结果表明用酵母双杂交一次筛选随机多肽文库既可以全面研究ErbinPDZ结构域的结合特性,又可以在全蛋白质组水平上找到表达于不同组织的天然配体。关键词:PDZErbin识别特性酵母双杂交随机多肽文库3中国协和医科大学博士学位论文Abstract:E3、rbin(ErbB2interactingprotein)playsacriticalroleinrestrictingErbB2tothelateralmembraneofhumanintestinalepithelialcellsinPDZdomaindependentmanner,BecauseofthepotentialtherapeuticvalueofdisruptingErbB2basolateraltargeting,weinitiallysetouttofurthercharacterizethebindingligandso4、fErbinPDZdomain.Yeasttwo-hybridscreeningofrandompeptidelibrarywasemployedhere,TheexperimentsshowedthatbesidesthereportedClassI·-[E/D]-·[T/S]·W-Vmotif.,ErbinPDZcanalsorecognizetheClassII—E一[V/A]-侈二扩andClass11I-D—E一肛矿motifs.Atthe一1position,acidicresidueswereobservedalthougharo5、maticresiduesWandFwerethepredominantselections.Interestingly,bothhi曲affinityligandscarryingtheC-terminalETWVandlowaffinityoneswithESDVwereidentifiedbyoncescreening.Alignmentofscreening-derivedligandsestablishedclearconsensussequencesattheCtermini.Weusedtheconsensussequencest6、osearchtheSwiss—ProtDatabasetoretrievethepotentialnativeligands.4outof13mostpromisingcandidateligandswereconfirmedinone‘to’oneyeasttwo—hybridexperiments.Thenovelinteractionsidentifiedheremayrevealsignificantcluesforfitrtherfunctionalinvestigation.Theresultssuggestedthatyeast7、two-hybirdscreeningofrandompeptidelibrarywasnotonlysufficientforbindingpropertycharacterizationofPDZdomains,butalsosuitableforpredictingphysiologicallyrelevantligandsforPDZdomains.Keywords:PDZdomainErbinBindingPropertyYeasttwo—hybridsystemRandomPeptideLibrary4中国协和医科大学博士学位论文前8、言PDZ(PSD一95,DLG—A,z_o一1首字母的缩写)结构域是一种广泛存在的蛋白相互作用结构域,SMART网站列出有1163个人PDZ结构域分布
