1、催产素预处理对胎鼠海马脑片缺氧无糖损伤后OTR、GABAARβ1表达的影响【摘要】目的:研究催产素(OT)预处理对胎鼠海马脑片缺氧无糖损伤后催产素受体(OTR)、γ氨基丁酸A受体β1亚基(GABAARβ1)表达的影响,探讨OT神经保护的作用机制。方法:采用胎龄20dSD大鼠海马脑片,实验组经含10-6mol·L-1OT人工脑脊液(ACSF)孵育预处理后行缺氧无糖损伤(OGD),对照组经正常ACSF孵育后直接行OGD,各15min,SP免疫组化法半定量测定海马CA1区OTR、GABAARβ1表达变化,电镜观察神经元损伤情况。结果:实验组海马CA1区OTR、GABA
2、ARβ1表达均显著高于对照组(t=2.118、P=0.048;t=2.272、P=0.036)。对照组大量神经元受损,核固缩或碎裂,部分核膜溶解,实验组细胞核、粗面内质网、线粒体均较对照组有所改善,核膜较光滑,胞浆肿胀较对照组减轻。结论:OT上调胎鼠海马OTR、GABAARβ1表达,减少神经元变性坏死,增强大脑耐受缺氧损伤的能力,对胎儿中枢神经系统起保护作用。【关键词】催产素;海马;缺氧无糖损伤;催产素受体;γ氨基丁酸A受体β1亚基 [Abstract]Objective:Tostudytheeffectsofoxytocin(OT)onchangesofox

5、expressions,andobserveneuronaldamageinelectronmicroscopy.Results:TheexpressionsofOTR,GABAARβ1inCA1areaofhippocampusweresignificantlyhigherintheexperimentalqroup(t=2.118,P=0.048;t=2.272,P=0.036).Alargenumberofdamagedneurons,nuclearcondensationorfragmentation,someofthenuclearmembranedis
6、solvedinthecontrolgroup;whilenuclei,roughendoplasmicreticulum,mitochondriawereimproved,nuclearmembranewassmooth,cytoplasmicswellingwereeasedintheexperimentalgroup.Conclusion:OTcanincreasehippocampalOTR,GABAARβ1expressions,decrease12neuronaldegenerationandnecrosis,enhancethecapacityoft
