2、itoperesearch,withanemphasislaidonthemostrecentachievements,i.e.thecorrelationbetweenantigenicityandpolypeptideprimarystructure.Theantigenicpropertiesofcoronavirusesresidemainlyintheirenvelopeglycoproteins:theS(spike)protein,ahighmolecularweightpolypeptidew
3、hichconstitutesthepetal-shapedpeplomersandtheM(membrane)protein,asmallerintegralmembranepolypeptide.Somecoronavirusespossessanadditionalsurfaceglyco•protein,thehaemagglutinin-esterase(HE)protein.PEPLOMERPROTEINSAnalysisofthepredictedaminoacidsequencesnowest
4、ablishedfor5coronaviruses(IBY,MHY,TGEY,FIPYandBCY)hasledtoamodelinwhichtheproteinisdividedinto2domains:theamino-terminalhalf,whichisassumedtoformtheglobularpartofthepeplomer,andthecarboxy-terminalhalf,whichisassumedtoformanelongatedstructure,thestalkofthepe
5、plomer,andalsocontainsaputativetransmembraneanchoringregionnearthecarboxyterminus.Bothpartsbearnumerousasparagine-linkedcomplexglycansidechains,whichmaymodulatetheirantigenicity.Thesequencesoftheamino-terminalhalvesexhibitverylittleconservation,whereashomol
6、ogiesaveraging30%areobservedinthecarboxy-terminalhalves.Severalcoronaviruses,includingmY,MHYandBCYhaveaSproteinwhichiscleavedapproximatelyinthemiddle,thusdefininganamino-terminalsubunit(SI)andacarboxy-terminalsubunit(S2).Severalmajorantigenicsiteshavebeenid
7、entifiedontheSproteinsofMHY,TGEYandmybycompetitivebindingexperiments;anumberofthesesitescouldbeoriented,ifnotpreciselylocalised,ontheprimarystructureofthemolecule.MHYSProteinRecombinantvirusesgeneratedinvivobyRNA-RNArecombinationwhichcontainapproximatelythe
8、5'two-thirdsoftheMHY-JHMpeplomergeneandthe3'one-thirdoftheMHY-A59peplomergenehavelosttheirreactivitytomonoclonalantibodies(MAbs)defmingthesitesdesignatedAandBontheJHMSprotein.Itwasinferredthatbothsites
