1、黄素对慢性应激大鼠肾上腺、血清ACTH和免疫功能的影响【摘要】目的:探讨姜黄素对慢性不可预知应激模型大鼠的肾上腺结构、血清促肾上腺皮质激素(ACTH)浓度和免疫功能的影响及其意义。方法:采用多种不可预知的刺激方式交替、持续应激20d建立大鼠慢性应激模型。随机分为正常对照组、应激模型组、姜黄素组(2.5mg/kg、5mg/kg、10mg/kg)、丙咪嗪组(10mg/kg),每组6只。采用HE染色技术,观察应激大鼠肾上腺结构的改变及姜黄素的作用。通过测定应激大鼠外周血中白细胞(WBC)数量和血清中ACTH和白细胞介素1β(Interleukin
2、-1β,IL-1β)含量的改变,观察姜黄素的作用。结果:慢性应激大鼠与正常对照组比较肾上腺皮质增厚,髓质萎缩,外周血白细胞数量明显减少(包括淋巴细胞、单核细胞和中性粒细胞百分比下降),血清中ACTH和IL-1β含量明显增加。姜黄素组大鼠肾上腺结构变化不明显,外周血WBC数量不同程度增加,血清中ACTH和IL-1β含量降低。结论:姜黄素可能通过调节下丘脑-垂体-肾上腺轴以及免疫系统功能,在慢性应激大鼠模型中显示出抗抑郁活性。【关键词】姜黄素应激抗抑郁肾上腺白细胞计数ACTHIL-1βAbstract:Objective:Toobserveth

4、ndimipraminegroup(10mg/kg).Andthenregularmethodwasusedtopreparepathologicsectionofadrenalgland,andbloodcellsanalyticsystemwasusedtodetectwhitebloodcell(WBC)count(includingrelativeWBCpercentages)inperipheralblood.TheserumlevelofACTHandIL-1βwerealsodetectedusingtheenzymelink
5、edimmunosorbentassay.Results:Chronicstressresultedindiffusehyperplasiaoftheadrenalcortexandatrophyoftheadrenalmedullainrats,andtheserumlevelofACTHwasincreased,whichsuggestedthatstress-adaptionfailureoftheadrenalglandoccurred,whileadrenalglandoftheratspretreatedwithcurcumin
6、(10mg/kg)priortoeachstressorfor21daysdidnotresultinanypathologicchangesandthelevelofACTHdecreased.Moreover,chronicstressalsosignificantlyreducedtheWBCcountandrelativeWBCpercentagesintheperipheralblood,includingthepercentageoflymphocytes,monocytes,neutrophils,however,curcum
7、inreversedthesechangesandraisedWBCcount,alongwithrelativeWBCpercentagessignificantly.Meanwhile,theserumlevelofIL-1βwasincreasedinchronicallystressedanimals,andcurcuminalsodeclinedthelevelofthisstresshormone.4Conclusion:Theseresultssuggestthatcurcuminexertsantidepressanteff
